Dopo essere stato per anni consulente per Scotland Yard in Gran Bretagna e dopo essere uscito da una clinica per disintossicarsi dall'alcool e dalle droghe, Sherlock Holmes si stabilisce negli Stati Uniti a New York City, dove accetta di collaborare con la polizia e risolvere diversi casi con l'aiuto della logica e del suo intuito. Il detective è affiancato, inizialmente suo malgrado, dall'ex chirurgo Joan Watson, la sua terapista di riabilitazione, che diventa in seguito sua assistente investigativa.
A coadiuvare le indagini per conto del New York City Police Department sono il capitano Thomas Gregson (ispirato all'ispettore Tobias Gregson che compare in alcuni libri di Arthur Conan Doyle) e il detective Marcus Bell.

Following his fall from grace in London and a stint in rehab, an eccentric modern Sherlock Holmes relocates to Manhattan, where his wealthy father forces him to live with a sober companion, Dr. Joan Watson. Formerly a successful surgeon until she lost a patient, Watson views her current job as another opportunity to help people. However, Sherlock is nothing like her previous clients. He informs her that none of her expertise as an addiction specialist applies to him and that he has devised his own post-rehab regimen – resuming his work as a police consultant in New York City. Watson has no choice but to accompany her irascible new charge on his jobs.
Over time, Sherlock finds her medical background helpful, and Watson realizes she has a knack for investigation. Sherlock’s police contact, Captain Thomas Gregson, knows from previous experience working with Scotland Yard that Sherlock is brilliant at solving cases, and welcomes him as part of the team. The investigative group also includes Detective Marcus Bell, an investigator with sharp intuition and formidable interrogation skills. Although initially skeptical of Holmes and his unorthodox methods, Bell begins to recognize Sherlock as an asset to their investigations.

But Sherlock finds her medical background helpful, and Watson realizes she has a knack for playing investigator.
With the mischievous Sherlock Holmes now running free in New York solving crimes, it’s simple deduction that he’s going to need someone to keep him grounded, and it’s elementary that it’s a job for Watson.
Amidst Sherlock and Joan’s battle with tech billionaire Odin Reichenbach, the investigators receive word of their old enemy and Sherlock’s former love, Jamie Moriarty.
Reichenbach inadvertently provides Sherlock and Joan with a lead that could give them evidence that brings him to justice.
Sherlock and Joan join forces with Holmes’ father, Morland to enlist his vast criminal network to help disassemble Reichenbach’s crime prevention system.
Holmes and Watson receive a shocking request to collaborate from their foe, Odin Reichenbach, when the tech billionaire asks them to test his new crime prevention system. Tasked with investigating someone the program predicts will soon commit a crime, the duo worries Odin will take drastic action before they conclude their inquiry.
Holmes and Watson work to determine if a long dormant serial killer has resurfaced in the wake of a sculptor’s murder in New York City. Also, Watson suspects Holmes is keeping something from her regarding tech billionaire Odin Reichenbach.
