The Upper East Side is thrown for a loop when Gossip Girl reports that Serena van der Woodsen (Blake Lively), Manhattan's most notorious party girl, has arrived back in town after a mysterious absence where no one knows where she was or why she left. Not even Serena's closest friend, Blair Waldorf (Leighton Meester), knew her whereabouts and is shocked to learn Serena has returned. However, their once close friendship is complicated upon Serena's return, strained now that Serena is the center of attention and Blair has been cast from the limelight, and the revelation that Serena had a fling with Blair's boyfriend Nate Archibald [(Chace Crawford).
Also ruling the Upper East Side is Chuck Bass (Ed Westwick), son of the wealthy and powerful Bart Bass and a friend of Nate's since childhood. Chuck leads a reckless life, which often leads Nate down a similar path of darkness and destruction.
The series begins with the return of Upper East Side "it girl", Serena van der Woodsen (Blake Lively) from a mysterious stay at a boarding school in Cornwall, Connecticut. Blair Waldorf (Leighton Meester), whom creators describe as the queen at the center of their chess game, is a longtime friend and occasional rival of Serena's, and the queen bee of Constance Billard School's social scene. The story also follows Chuck Bass (Ed Westwick), the bad boy of the Upper East Side; "golden boy" Nate Archibald (Chace Crawford), Chuck's best friend and Blair's boyfriend for many years. However, their relationship had been rocky ever since Serena left for boarding school. Other characters of the turbulent Manhattan scene: Dan Humphrey (Penn Badgley), Dan's best friend Vanessa Abrams (Jessica Szohr), and Dan's sister, Jenny Humphrey (Taylor Momsen).

After a run-in with an increasingly paranoid Bart, Blair grows alarmingly concerned for Chuck’s safety. However, feeling confident that his father will never hurt him, Chuck makes a deal that could change his life. Wanting to make things better for Chuck, Blair devises a plan with the help of Serena, Georgina and Sage to try and get what they need out of Bart. Meanwhile, Nate's financial dilemma finally lands him in a place that he never thought he would be.
Serena and Dan decide to throw their first Thanksgiving together, inviting only their closest friends, but everyone seems to have their own agenda for attending. Feeling defeated by his father, Chuck begins a downward spiral, leaving Blair to find a solution to his problems. Meanwhile, Dan has written his latest exposé, but he must make a tough decision whether to publish it, knowing it could destroy a close friendship.
Serena and Dan realize that they need to make amends with the people they have wronged. Blair has one last chance to prove to her mother her worth as a fashion designer and with Serena’s help, comes up with a plan to make her line the “it” look for Sage and her friends. Chuck is close to getting the evidence he needs against his father, until one of his closest allies reveals his plan to Bart.
Blair gets ready for her mothers imminent arrival, but is unprepared for the set of demands her mother puts upon her. Serena volunteers to help Dan find a new place to live, but their time together brings up unexpected emotions. With Ivy’s help, Chuck discovers there is evidence incriminating his father in illegal activities and enlists the help of Nate to retrieve it. Meanwhile, Rufus and Ivy are excited about their art gallery opening until they find an empty RSVP list and must scramble to find another way to make the event a success.
